Õppeaine eesmärgiks on anda laiapõhjaline ülevaade sellest mis on struktuuribioloogia, milleks seda vaja on ning anda ülevaade põhilistest väljakutsetest struktuuribioloogias. Kursuse lõppedes on tudengitel arusaam nii struktuuribioloogia põhilistest teoreetilistest alustest kui ka praktilistest väljunditest.

The aim of the course is to give an overview of what is structural biology, what is structural biology useful for and what are the main challenges in structural biology. By the end of the course the students will have basic understanding of the main theoretical aspects pertaining to structural biology as well as the practical applications of the discipline.

Üliõpilane saab aru makromolekulide kristalliseerimise, difraktsioonimustrite ning elektrontiheduskaartide interpreteerimise põhimõtetest. Tudeng oskab kursuse lõpuks lahendada röntgenkristallstruktuuri molekulaarse asenduse meetodi abil.

The student will understand the main principles of macromolecular crystallization, as well as the interpretation of diffraction patterns and electron density maps. Students will learn how to solve an X-ray crystal using the molecular replacement method.

Struktuuribioloogia on mõiste, mille alla saab liigitada terve rea biofüüsikalisi meetodeid bioloogiliste markomolekulide uurimiseks. Kursuse jooksul antakse ülevaade röntgenkristallograafiast ning röntgendifraktsiooni ja väikese nurga hajumise füüsikast. Ühtlasi käsitletakse kursusel bioloogiliste molekulide kristalliseerimise metoodikaid, difraktsiooni mustrite analüüsi, elektrontihedukaartide interpreteerimist ning peatutakse valkude röntgenstruktuuri lahendamisel. Antakse ka põgus ülevaade krüoelektronmikroskoopiast. Struktuuribioloogia kursuse vältel räägitakse jooksvalt ka olulistest verstapostidest molekulaarbioloogias ja biokeemias, milleni on jõutud just tänu struktuuribioloogiale.
brief description of the course in English
Structural biology is a term that can be used to describe a wide array of biophysical methods to study biological macromolecules. The course gives an overview of X-ray crystallography, X-ray diffraction as well as small angle X-ray scattering physics. The course will cover methods in biological macromolecule crystallization, analysis of diffraction patterns, interpretation of electron density maps, as well as the process of arriving at a structure solution. Small overview of cryo-electronmicroscopy will be given. Throughout the course, important land-mark discoveries in molecular biology and biochemistry will be discussed that were achieved due to structural biology methods.
